Osteomodulin impacts positively the bone remodeling process in osteoarthritis

چکیده
Purpose: Osteomodulin (OMD) is a Small Leucine Rich Proteoglycan (SLRP) highly specific of the bone extracellular matrix. Its production and expression are impaired in subchondral sclerosis associated with osteoarthritis (OA). This study aimed to investigate effects OMD on osteoblasts osteoclasts culture. Methods: Different concentrations (2.5, 10, 40 160 ng/ml), corresponding range concentration found supernatant primary vitro human osteoblast cultures, were tested sclerotic The from either or non-sclerotic area cortical OA patients isolated, serving as control. mineralization cultures was assessed Alizarin red staining at 17-day differentiation. To analyze effect osteoclasts, marrow mice harvested cells differentiated RANKL M-CSF into stopped 4-day osteoclast differentiation, (10 40ng/ml 5μg/ml) mixed M-CSF. number reported after TRAP staining. binding RANKL-OMD evaluated Solid Phase Binding Assay. In this test, coated Nickel plate, incubated OMD/RANKL complexes revealed polyclonal antibody. Results: No mortality observed tested. Sclerotic showed strong decrease comparison osteoblasts. partially restored (Fig. 1A). solid phase assay demonstrated ability bind RANKL. biological consequence murine Preliminary results pre-mix RANKL/M-CSF OMD, drastic it even fully blocked 5μg/ml 1B) Conclusions: major proteoglycan matrix turnover. It improves inhibits differentiation through capture content could trigger by initiating enhancing imbalance remodeling. research funded F.R.S.-FNRS for “EOS: Excellence Science -Join-t-against-Osteoarthritis”, grant 30480119 , Osteoarthritis Foundation www.osteoarthritisfoundation.eu .

Epigenetic Regulation of Bone Remodeling and Its Impacts in Osteoporosis
Epigenetics describes mechanisms which control gene expression and cellular processes without changing the DNA sequence. The main mechanisms in epigenetics are DNA methylation in CpG-rich promoters, histone modifications and non-coding RNAs (ncRNAs). DNA methylation modifies the function of the DNA and correlates with gene silencing. Histone modifications including acetylation/deacetylation and...
متن کاملSclerostin Promotes Bone Remodeling in the Process of Tooth Movement
Tooth movement is a biological process of bone remodeling induced by mechanical force. Sclerostin secreted by osteocytes is mechanosensory and important in bone remodeling. However, little is known regarding the role of sclerostin in tooth movement. In this study, models of experimental tooth movement were established in rats and mice. Sclerostin expression was investigated with immunohistochem...
متن کاملHalofuginone Attenuates Osteoarthritis by Rescuing Bone Remodeling in Subchondral Bone Through Oral Gavage
Osteoarthritis (OA) is a common debilitating joint disorder worldwide without effective medical therapy. Articular cartilage and subchondral bone act in concert as a functional unit with the onset of OA. Halofuginone is an analog of the alkaloid febrifugine extracted from the plant Dichroa febrifuga, which has been demonstrated to exert inhibition of SMAD 2/3 phosphorylation downstream of the T...
متن کاملPhysiological bases of bone regeneration II. The remodeling process.
Bone remodeling is the restructuring process of existing bone, which is in constant resorption and formation. Under normal conditions, this balanced process allows the renewal of 5-10% of bone volume per year. At the microscopic level, bone remodeling is produced in basic multicellular units, where osteoclasts resorb a certain quantity of bone and osteoblasts form the osteoid matrix and mineral...
متن کاملConnecting Mechanics and Bone Cell Activities in the Bone Remodeling Process: An Integrated Finite Element Modeling
Bone adaptation occurs as a response to external loadings and involves bone resorption by osteoclasts followed by the formation of new bone by osteoblasts. It is directly triggered by the transduction phase by osteocytes embedded within the bone matrix. The bone remodeling process is governed by the interactions between osteoblasts and osteoclasts through the expression of several autocrine and...
